De prognose van patiënten met een recidief van een primair centraalzenuwstelsellymfoom (PCZSL) of een CZS-recidief van een grootcellig B-cellymfoom (sCZSL) is uitermate somber. Golcadomide is een nieuwe cereblon-E3-ligasemodulator (CELMoD) gericht op de afbraak van transcriptiefactoren Ikaros en Aiolos, die onder andere via downregulatie van cMYC en IRF-4 leidt tot celdood van lymfoomcellen. Bij recidief DLBCL zien de resultaten van de fase I/II-studie er veelbelovend uit. Golcadomide heeft een goede passage van de bloed-hersenbarrière. In de HOVON 176-studie zal de effectiviteit van golcadomide worden onderzocht bij patiënten met een recidief PCZSL en sCZSL.
